Of all the hotels I've had the pleasure of visiting, the most incredible, to date, has been the Venetian in Las Vegas, Nevada.  The Guinness Book of World Records states that the Venetian has the largest rooms in the world.  I would add that they are also the most beautifully appointed rooms that I've ever stayed in.  And I only stayed in one of the "standard" rooms at the hotel (referred to as "Luxury Suites").
